in the following diagram, the server motherboard is connected to four M.2 SSDs (4x) through CEM2SLIMSAS-EVM and DS160PR410EVM-RSC. If the PCIe x16 link of the server motherboard is one link, does the x16 link of the server motherboard have to support splitting? Do I have to set x4x4x4x4 in the BIOS of the motherboard?

Yes, the CPU and BIOS of the system would need to support x4x4x4x4 bifurcation.
The DS160PR410 devices inherently support bifurcation, but do not play any role in determining the PCIe link's bifurcation settings.
